Heraklion

You'll see diferent spellings of this place, but it all means the same thing.  This is the main city on Crete and most likely your point of entry, be it by ferry or plane.  The main thing to see here is the Archeological Museum and after that you're probably better off exploring the rest of the island.

The museum is located on the main square and is open 8-7 (not open Monday morning) in the summer and 8-5 during the winter.  It costs between 5 and 10 Euro, as I recall.  So what are the cool things to see here?  There are the bull symbols that Minoan culture is known for are all on display.  This most likely started the Minotaur stories of Ancient Greece.  Be sure to stop by the snake goddess statue found at Knossos, a famous representation of the Minoan diety.  Upstairs are more treasures with the frescoes found on the walls at Knossos.

The museum can get crowded and full of tours, so get there when they open or right before they close if possible.
